Abstract Transitioning from petrochemical-based to microbial-based processes requires robust biocatalysts for chemical production. Biofilms play a crucial role in the development of microbial technologies for wastewater and gas treatment, bioremediation, microbial electrosynthesis, and chemical production, as they can attain high cell density, tolerate process stressors, and support continuous operation. Here, we report a drip-flow reactor in which recombinant Pseudomonas taiwanensis VLB120 forms biofilms on a porous poplar- polyethylene terephthalate (PET) material to develop hybrid living materials for continuous biotransformation of cyclohexanol to adipic acid. Biofilm growth over 7 days was examined under different cultivation conditions, using a design of experiments (DoE) approach to establish a biofilm with an approximately 300 gBDW m−2 biomass density. In the biotransformation phase, a dynamic glucose feeding approach enabled stable conversion of 10 mM cyclohexanol to adipic acid over several weeks. Our work demonstrates the resource-efficient development of hybrid living materials to maximize biotransformation performance for adipic acid production.
