Abstract Biota samples represent one of the most challenging matrices to handle regarding organic micropollutant analysis due to complex matrix constituents including different lipids and proteins amongst others. In this chapter, a broad overview of the state-of-the-art sample preparation methods and their most recent applications including respective advantages and limitations is presented for multi-residue analysis. The advancements in analytical instrumentation and methods over the last decades have opened new possibilities and a better understanding of the monitoring of organic micropollutants in biota. Research interest has shifted from developing specific methods for a small number of hydrophobic organic contaminants with similar properties to multi-residue target screening methods for a wide range of chemicals with diverse properties including chemicals of emerging concern. High-resolution mass spectrometers preceded by chromatographic separation have become one of the main instrumentations hence opening up for suspect and nontarget screening in biomonitoring studies. All these developments require a change in sample handling strategies for biota samples. Considering these aspects, with this chapter, we aim to provide a guidance document for researchers working with biota samples to make the most of their samples and existing instrumentation.
