Abstract | Effect-directed analysis (EDA) is an approach to identify chemicals in complex mixtures which exert adverse effects. To this end, a combination of bioassays for effect detection, fractionation to reduce sample complexity, and chemical analysis mainly by liquid or gas chromatography coupled to mass spectrometry is used. High-resolution mass spectrometry (HRMS) using orbitrap or time-of-flight analysers has become a key technique for EDA, as it allows for an equally sensitive and selective analysis of samples without a pre-definition of the compounds of interest. This chapter provides an overview on the state of the art of HRMS in EDA studies on water and sediment. The main HRMS-based strategies such as target screening, screening for suspected toxicants, library search, and true unknown identification are presented, highlighting the way these were used in different studies including their strengths and current limitations. |
