| Abstract | This critical review addresses the widespread contamination of terrestrial and aquatic ecosystems by pharmaceutical active compounds (PhACs), their transformation products (TPs), and microbial contaminants (including antimicrobial resistance and pathogens), with the aim of identifying key conceptual and methodological priorities for enhancing environmental risk assessment (ERA) through improved consideration of environmental microbial communities. Working up from a One Health perspective, this paper synthesizes current knowledge, highlights major gaps, and outlines strategies for tackling six key challenges tied to three core scientific questions: (i) what is the occurrence, dynamics, and persistence of PhACs, their TPs and microbial contaminants within soil and aquatic environments? (ii) how does interaction between environmental microbial communities and PhACs, their TPs and microbial contaminants affect soil and aquatic ecosystem processes and the environmental fate of these substances? (iii) how can microorganisms be more effectively integrated into ERA frameworks and regulatory strategies? We outline research directions in the field and the supporting technical tools and approaches to mobilize, which include big data-driven approaches (e.g. non-target chemical screening, meta-multiomics), hypothesis-driven experiments and modeling, effect-based assessments (e.g. effect-directed analysis: EDA), and in silico methods (e.g. quantitative structure–activity relationship models: QSARs). We underline the importance of open science and data sharing to enhance the availability of ecotoxicological data, improve environmental quality standards, and strengthen ERA. We further identify the main barriers associated with the six challenges we propose and highlight how addressing them could make important contributions to environmental management by advancing ERA and supporting the development of mitigation strategies. |
